I would try setting the usb to com 1 and see if it works on that.

If you go to Control Panel>System>Hardware Tab>Device Manager>Ports (Com & LPT)> +, you should see your device listed.

Double click 'Prolific USB.....>Port Settings Tab>Advanced> Change drop down to 'COM 1'
Click OK.

Now try DMLoader on COM1.

If that doesn't work, I can only think that you have a hardware conflict.
